Firstly, $2 will be the automatically created name for this new constraint.
Secondly || doesn't mean OR - it joins strings.
Try something like (untested):
ALTER TABLE genus ADD CONSTRAINT valid_gender CHECK (gender IN
('masculine','feminine'));
